PFU/mL = plaques × dilution factorstep ÷ volume plated (mL). Counts outside the countable window (30–300 per plate, 3–30 per spot) are flagged but still included — tick Excl to drop a replicate from the stats, or TNTC when a plate was too numerous to count. A spot assay plates far less than a full plate — set the volume to the drop you actually spotted (often 5–10 µL), not 100.
